Regulation of Granulosa Cell Proliferation by Growth Factors in Hens.

The aim of this study was to determine if the granulosa cells produce the growth promoting factor(s) which regulate their own proliferation. Conditioned medium from unstimulated granulosa cells (GCCM) and control medium for it (CFCM) were prepared by two days culture of the granulosa cells from the second largest follicle and by an incubation of the identical medium without cells. Conditioned medium from dibutyryl cyclic AMP (dbcAMP) stimulated-granulosa cells (cAMP-GCCM) and control medium for it (cAMP-CFCM) were prepared by two days culture of the granulosa cells in the medium containing 1mM dbcAMP and by an incubation of the identical medium without cells. Newly isolated granulosa cells were cultured in these conditioned media, and after 6 days culture the cell number was counted. The difference in the cell number was not significant between the cultures of GCCM and CFCM. However, the number of granulosa cells grown in cAMP-GCCM was significantly greater than that of the cells grown in cAMP-CFCM. These results suggest that growth promoting factor(s) are produced by the granulosa cells under the stimulation of cAMP, and this factor may be responsible for the process of the granulosa cell proliferation during the follicular growth.
