Effect of insemination dose and re-dilution rate on the fertility of fowl semen frozen in Hiroshima diluent.

In order to find the optimum conditions for efficient use of fowl frozen semen, the experiments were conducted to determine the minimum dose of insemination and maximum rate of dilution at redilution after freeze-thaw. Semen were frozen in pellets in the Hiroshima diluent. After centrifuged the freeze-thaw semen spermatozoa in freeze-thaw semen was rediluted to the same concentration as in the fresh semen.1. The fertility during the period from 2 to 8 day after single insemination dose of 0.025ml, 0.05ml, 0.1ml and 0.2ml of freeze-thaw semen was 60.9, 90.4, 96.9 and 94.6%, respectively.2. The fertility during the period from 2 to 8 day after single insemination dose of 0.2ml of freeze-thaw semen further diluted 1-fold, 2-fold, 4-fold and 8-fold with re-diluent was 94.6, 80.0, 82.7 and 39.7%, respectively.3. When the number of spermatozoa deposited were adjusted to approximately 10 million, 60.9, 67.1, 33.1 and 39.7% of eggs were fertile during the period from 2 to 8 day after single insemination dose of 0.025ml semen diluted 1-fold, 0.05ml semen diluted 2-fold, 0.1ml semen diluted 4-fold and 0.2ml semen diluted 8-fold, respectively.These results indicate that insemination dose of more than 0.05ml of original sperm concentration is required when high fertility levels (>90%) are expected and that the deposition volume of 0.05ml of semen diluted 2-fold is most efficient in order to obtain the maximum number of fertilized eggs per semen.
