The usage of specific pathogen free (SPF) eggs is desirable to produce some avian live virus vaccines and to make quality control of vaccines. The reserch was performed to evaluate a system and facilities for production of SPF eggs in Kitasato University, Yakumo Farm, Hokkaido.The SPF chickens were kept in filtered air under positive pressure (FAPP) type house A and B at 13-25°C of environmental temperature. House A for production of SPF eggs has 748 m2 of floor space in total; 304 m2 for 1st floor, 244 m2 for 2nd floor and 200 m2 for the underground stockroom, and about 1600 layers are kept in cages or slatted floor feeding system. House B has 405 m2 floor space for the maintenance stock of about 250 SPF chickens in cage systems.All materials and facilities to be brought into the chicken house are disinfected by the use of ethylene oxide gas, autoclaving or formaldehyde fumigation. The chicken house is disinfected according to the standard procedure used in the windowless chicken house. The germfree condition in SPF area is confirmed by measuring number of viable bacteria using stamp agar method. SPF chickens are checked on 18 diseases according to the method determined by the Committee of Avian Live Vaccine, Japan at the SPF Inspection Center of the Kitasato University. All the works on the production of SPF eggs in this institution are operated by 3 persons.
