Effect of medium pH on the growth and ascus formation in dermatophytes.

Various strains of Trichophyton mentagrophytes, T. rubrum, Epidermophyton floccosum, and Nannizzia grubyia were examined for their growth and conidial production on two media, Sabouraud dextrose agar (SDA) and diluted SDA at pH ranges from 3.5 to 10. The pH value was determined using a pH meter (Beckman-Toshiba LAB-O-MATE) with electrode (COMB pH Electrode 39507). The fungi were cultured at 25°C for 3 weeks. Additional mating experiments of N. grubyia in defferent pH media were also carried out.The range of pH values for growth did not differ between the two media. The range compatible with growth was 3.5 to 10 in T. mentagrophytes, T. rubrum, and N. grubyia and 4 to 10 in E. floccosum. The most rapid growth rate was observed during the second week after inoculation. Conidial formation and pigment production in most dermatophytes were specific to each strain and did not depend on the pH value of the media.Increased ascospore formation of N. grubyia was observed on diluted SDA at pH values from 4 to 9 and the mating was most successful at a pH of 7.
