Effect of bifemelane and .ALPHA.-tocopherol on the brain edema in focal cerebral ischemia in rats.

Focal cerebral ischemia caused by occlusion of the middle cerebral artery (MCA) was induced in Wistar rats.<BR>Brain tissue free fatty acids (palmitic, stearic, oleic and arachidonic acids), arachidonic acid metabolites (prostaglandin E<SUB>2</SUB>, 6-keto-prostaglandin F<SUB>1</SUB>α, thromboxane B<SUB>2</SUB> and leukotriene C<SUB>4</SUB>), energy metabolites (AMP, ADP, ATP and lactate), and water content of the occluded hemisphere were determined before ischemia and 6 hours after occlusion of the middle cerebral artery. The effect of bifemelane and/or α-tocopherol on changes of free fatty acids, arachidonic acid metabolites, energy metabolites and water content was evaluated. Bifemelane (25 mg/kg, 30 minutes before MCA occlusion) and α-tocopherol (20 mg/kg, 60 and 30 minutes before MCA occlusion) were given intraperitoneally.<BR>Total free fatty acid levels (sum of 4 fatty acids) increased significantly after MCA occlusion, but bifemelane and/or α-tocopherol did not have any effect on free fatty acid levels. Leukotriene C<SUB>4</SUB> and thromboxane B<SUB>2</SUB> increased significantly after MCA occlusion. Bifemelane and/or α-tocopherol reduced leukotriene C<SUB>4</SUB> accumulation, but arachidonic acid metabolites of the cyclo-oxygenase pathway failed to show any reduction by bifemelane and/or α-tocopherol pretreament. ATP and energy charge reduced and lactate increased significantly after MCA occlusion. Bifemelane and/or α-tocopherol pretreatment did not have any effect on changes of energy metabolites. Brain water content significantly increased after ischemia compared to the controls. Pretreatment with bifemelane and α-tocopherol inhibited the increase of brain water content in the occluded hemisphere. Thus, bifemelane and α-tocopherol inhibited the development of ischemic brain edema.<BR>The anti-edema effect of pretreatment with bifemelane and α-tocopherol seemed to be due to both the inhibition of lipoxygenase activity and the anti-oxidant effect.
