Quantitation of Cerebrospinal Fluid Immunoglobulins

Cerebrospinal fluid (CSF) immunoglobulins are quantitated by electroimmunodiffusion and the antibody-agar method. For CSF IgG quantitation by the former using immunoelectrophoresis apparatus (20×30×8cm), the best conditions of the volume of sample, antiserum (Behringwerke), electrophoresis voltage and time are 2 μl, 0.2ml, 150 voltage and 300 minutes, respectively. Under these experimental conditions, CSF IgG content of 14 non-neurological persons ranges from 2.5 to 6.3mg/dl with an average of 4. 1mg/dl. The normal range of CSF IgG/total protein ratio is 5. 0-12. 6%. Although the number of cases is so far very limited, the ratio is high in two of the two cases with retrobulbar neuritis and two of the three cases with multiple sclerosis and low in one case with myotonic dystrophy.<BR>CSF IgM and CSF IgA quantitation are also possible, although the values are much smaller than in case of CSF IgG. In one case with meningioma, CSF IgA is counted to be 5mg/dl.<BR>CSF immunoglobulin quantitation by the antibody-agar method is also possible. Standard curve for CSF IgG and IgM measurement and an example of CSF IgA quantitation are shown.
