The determination of serum glucose with o-toluidine using a glacial acetic acid medium is sensitive, precise and accurate enough for the requirements of a clinical laboratory. It is not convenient for a routine work because g acial acetic acid has irritant vapor.<BR>Therefore, a new method of determination of serum glucose with o-toluidine using no glacial acetic acid (TGBM method) is presented. The procedure employs a mixture of glycollic acid, benzyl alcohol and methyl cellosolve instead of glacial acetic acid.<BR>The new reagent (TGBM reagent) is non irritant, colorless, and stable for at least several months when stored at 4° in a brown bottle.<BR>The coefficient of variation was ±2% by the TGBM method; recovery experiments gave a mean yield of 100%. Good agreement was observed between the TGBM method and the UV method using hexokinase-G-6 PDH or OTB method.
