Simultaneous Determination of Cortisol and Cortisone in Human Saliva Using Semi-Microcolumn:Liquid Chromatography with Column-Switching System and Ultraviolet Detection

A column-switching system based on a semimicrocolumn with ultraviolet detection for simultaneous direct analysis of cortisol and cortisone in human saliva samples was developed. The characteristics of this method are that it allows simultaneous determination of cortisol and cortisone with ultraviolet detection by direct analysis of human saliva samples, and that the amount of the mobile phase used about one-tenth in comparison to that in the conventional method. The recoveries of cortisol and cortisone were 99.5-103.9% and 98.7-105.1%, respectively. Errors in the reproducibilities of these results were both less than 3%. The detection limits for cortisol and cortisone were 0.3pmol/ml each. Circadian rhythm was observed using the present method, and there was a good correlation between the concentration of cortisol and that of cortisone in human saliva samples (r=0.946, n=48), and between the present method and the RIA method for cortisol (r=0.913, n=48).
