As previously reported, a preliminary experiment was set up in order to run a comparision among various methods using trihydroxyindole reaction with respect to the recovery, accuracy and rapidity. The present work deals with an improvement of Price's method for simultaneous determination of epinephrine and norepinephrine in human plasma. The improvements were as follows.
1) Purification of acid washed aluminum oxide (Merck #71695 acid washed)
2) Addition of a large amount of EDTA disodium.
3) Prevention of change in fluorescence intensity arising from progressive decay of fluorescence of trihydroxyindole compounds formed.
Thus, the determination method was considerably improved in accuracy and rapidity. Using this method, it was shown that while in normal subjects almost no plasma catecholamine was detected, in the patients investigated sporadic detection was made in some diseases, in which catecholamine detection hitherto may have been difficult.
