New resin-based supports for HPLC to separate proteins by hydrophobic interaction

New resin-based supports of various hydrophobic functional groups such as phenyl, butyl and ether were prepared for HPLC of proteins and evaluated under each elution conditions. Commercially available proteins were used as samples and eluted without the interaction with the supports in case of stronger anions of chaotropic effect. Effect pf kinds of gradient salts on the separation performance was studied. Retention times decreased in the order of Hofmeister′s series for cations and anions of the salts. For acidic or neutral proteins, a linear relationship was observed between logarism of the molecular weight and the retention time. Hydrophobicities of the proteins were also correlated with the retention times. Thus, hydrophobic separation conditions of representative proteins on the three columns were obtained. The columns will be useful for biological samples.
