Synthetic luteinizing hormone-releasing hormone (LH-RH) was administered to pregnant Wistar rats for the examination of delivery and pre-postnatal development of their offspring. The compound was given to the rats intravenously, in doses of 6, 15, 30 600 and 6000 μg/kg/day, for 6 days from day 9 to day 14 of gestation.<BR>The results are summarized as follows : <BR>(1) No significant toxicity was observed in the rat mothers during the medication period. Body weight gain of the mothers were comparable for all groups.<BR>(2) Autopsy of the full-term mothers revealed an increase in the number of corpola lutea at doses of 60,600 and 6000 μg/kg.<BR>(3) Intrauterine mortalities and mean body weights of fetuses are considered to be unaffected by the treatment at any dosage.<BR>(4) Among live fetuses there were no malformations attributable to the LH-RH treatment, nor any tendency toward masculinization.<BR>(5) Some of mothers receiving 15 and 30 μg/kg of LH-RH had the onset of labor delayed for a few days. At the doses of 60,600 and 6000 μg/kg, the delays in paturition were more remarkable. Almost half of the mothers in these groups left fetuses in utero, and resulted in death by bacterial infection.<BR>(6) As for the normally delivered offsprings, both the postnatal development and sexual maturation were quite satisfactory.<BR>(7) The vialbility of the fetuses delivered from the LH-RH treated mothers by cesarotomy was confirmed by foster nursing.<BR>From the facts obtained in the present study, we may conclude as follows : Synthetic LH-RH has no adverse effects on the pre- and postnatal development of rat fetuses, but a delaying effect on parturition is evident with daily doses of more than 15 μg/kg.
