Angiotensin II has been shown to induce aldosterone synthesis in man, and to stimulate cortisol as well as aldosterone synthesis in isolated bovine adrenals.<BR>The present study has been carried out to determine whether low concentration of angiotensin II stimulates 11-OHCS production in isolated bovine adrenal cells, and whether steroidogenesis by angiotensin II can be utilized for the bioassay of angiotensin II.<BR>Isolated cells were prepared by a modification of the Hanig procedure from bovine adrenal inner slices. These cells responded to the addition of valyl-5-angiotensin H by increased production of 11-OHCS. And 11-OHCS levels were determined by a modification of the Silber procedure.<BR>Production of 11-OHCS by the isolated cells in response to angiotensin II was increased in parallel with calcium and potassium concentrations of the incubation medium. Net production of 11-OHCS was maximum at the calcium and potassium concentrations 5.08 mM and 8 mM respectively. Addition of bovine serum albumin (BSA) to the medium increased 11-OHCS production, and it was maximum at the BSA concentration of 0.75%.<BR>Minimum effective dose of angiotensin II for the stimulation of 11-0HCS production was 0.5ng. (p<0.01). The isolated adrenal cell method is of satisfactory accuracy (λ=0.038) for the dose range 5ng-20ng, where response is linearly related to the log of the dose.
