The hydration of proteins in erythrocytes was quantitatively investigated by the proton-proton nuclear Overhauser enhancement. It was found that the bound water fractions amount 0.37 g of water per g of protein, and have microviscosities at least 1000 times the bulk water viscosity. The use of Mooney's equation with hard spherical particels showed that the reported data for the intracellular viscosities are consistent with the viewpoint that the effective volume is given by the protein volume and the bound water volume. This finding suggests that the intrinsic viscosity is about 2.9 cm3/g.
