Estrogen metabolites were analized in urine of rabbits by chromatographic techniques after subcutaneous administration of such estrogens as estriol, estradiol-178, estradiol-17α or estrone.<BR>The results obtained were as follows : <BR>1. Estradiol-17α was the chief metabolite in urine of rabbits receiving injections of either estradiol-17β, estradiol-17α or estrone.<BR>2. Estriol, estradiol-17β, 16-hydroxyestrones and 2-metoxyestrone could not be detected in urine of rabbits treated with one of these estrogens (estradiol-17β, estradiol-17α or estrone) by the hydroquinone-Kober method.<BR>3. Small amounts of estrone and 17-epiestriol were noted in urine of the estrogentreated rabbit.<BR>4. Estriol was found to be partially converted to 17-epiestriol.<BR>5. As estradiol-17α was partially changed to unidentified metabolites by hot hydrochloric acid treatment, hot-acid-hydrolysis should not be used to hydrolyze conjugated estrogen in the rabbit urine.<BR>6. Metabolism of injected estradiol-17β was affected by liver damage induced experimentally : Central injury of liver lobules with carbon tetrachloride impaired neither the conjugation of estrogen with glucuronic acid nor the conversion to estradiol-17α, Peripheral injury of liver lobules with allyl formate greatly impaired the conjugation, but did not affect the conversion.
