The most difficult methodological problem in the determination for urinary estrogens is the interferring urochromogens which interfere with colour development, especially in low estrogen concentration.
Many attempts have been proposed to exclude this troublesome urochromogens. However, unfortunately, the problem is not completely solved.
Salokangas & Bulbrook reported the new method of estimation for urinary estrogens consists of Brown's method, and extraction procedure for Kober colours with tetrachloroethane (T.C.E.) containing p-nitrophenol.
The procedure of extraction for Kober colours was investigated and the following results were obtained :
1) Tetrachloroethane containing p-nitrophenol was the most favourable extractant for extraction of Kober colours.
2) The fading of extracted Kober colours was slight and negligible when placed in the dark and kept cool.
3) After the extraction, the maximal absorption was transmitted to a longer wave region from 516 mμ to 539 mμ, and the urochromogens were reduced remarkably.
4) By the reduction of urochromogens which appeared in a shorter wave region, the absroption spectra became sharp after the extraction.
5) In comparison with urinary estrogens, estimated by Brown's method and the extraction, good agreement was observed in estrone fractions, but was not so well cor-related in estradiol-17 β and estriol fractions.
6) The reasons for this discrepancy are discussed.
