Raben type human growth hormone (HGH) was immunologically studied, and then haemagglutination inhibition technique was employed to assay serum HGH. The levels of serum HGH were observed as they changed with age and with various kinds of endocrine or metabolic disorders. I. Immunological studies of HGH.<BR>Anti-HGH serum was prepared by the injection of Raben HGH into rabbits. Their immunological characteristics were investigated and the following results were obtained : <BR>1) The antigenicity of HGH was demonstrated utilizing Ouchterlony agar gel precipitation procedure, precipitin ring test, complement fixation reaction and haemagglutination reaction.<BR>2) It was recogized by the Ouchterlony procedure that Raben HGH had a protein contaminant, but the amount was so small that the anti-serum, which the author used, could be utilized to immunoassay serum HGH by the haemagglutination inhibition technique.<BR>3) In agar gel, anti-HGH serum did not react to human serum and pure human albumin, whereas a single band of precipitation formed with the crude human and monkey pituitary extract fused with that of HGH.<BR>4) Among various kinds of immunological methods such as Ouchterlony agar gel diffusion, precipitin ring test, complement fixation method and haemagglutination inhibition reaction, the most sensitive method to detect an extremely small quantity of HGH in human serum was the haemagglutination inhibition reaction.<BR>II. Serum HGH levels.<BR>Serum HGH of normal individuals and patients with various kinds of diseases was assayed with the haemagglutination inhibition technique, and the following results were obtained : <BR>1) Normal children and adults.<BR>Eighty-three normal individuals, including infants (5 to 12 months), 11 babies each aged 1 year, 15 children aged 2 to 4, 15 aged 5 to 7, 11 aged 8 to 10, 15 aged 11 to 15 and 9 adults, showed the following levels of serum HGH : <BR>a) There was no significant difference among the group saged 5 months to 7 years, and the mean value was 425±18 (S.E.) mμg/ml. For those over 8 years old, the serum HGH levels decreased and no significant diffnrence was observed in the groups aged 8 to 15. The average value in this group was 266±13 (S.E.) mμg/ml. Nine normal adults averaged 279±20 (S.E.) mμg/ml. Between female and male no significant difference was observed.<BR>b) The limit of rejection at 5% risk in the younger group (aged 5 months to 7 years) was 425±245mμg/ml, whereas that in the older group (aged 8-15) and in adults were 269±140mμg/ml and 279±147 mμg/ml respectively.<BR>2) Premature infants.<BR>The mean value in 9 premature infants without any complications was 187±25 (S.E.) mμg/ml, therefore the value was significantly lower than that in the normal children and adults.
