In order to deciding the subjects of diabetogenic or antidiabetogenic action of the pituitary and adrenocortical hormones, it should be important to confirm the action on ketosis of these hormones. These has been but little information available regarding on this problem and also the effects of these hormones on ketogenic activity are reported with conflicting results.<BR>The present investigation is undertaken to determine the changes of blood ketone bodies of alloxan diabetic rabbits or dogs after single injection of ACTH with or without butyrate and single injection of cortisone or DOCA, and also to find the effects of these hormones by their daily administration for a prolonged period.<BR>The following results have been obtained.<BR>I. Ketogenic activity of ACTH for alloxan diabetic rabbits.<BR>(1) In normal rabbits increase of the total ketone bodies in blood following the injection of ACTH was extremely small, however in alloxan diabetic rabbits, the total blood ketone bodies showed particular elevation of β-hydroxybutyric acid, and the increase was somewhat pronounced.<BR>(2) With the animals injected of sodium butyrate, the ketogenic influence reaches well marked degree and lasting a longer hours in alloxan diabetic rabbits in contrast to the healthy ones. While, a rise of acetone was almost parallel with β-hydroxybutyric acid in healthy animals, b It with the alloxan rabbits β-hydroxybutyric acid rose markedly and was followed with gradual depletion.<BR>(3) When ACTH was injected into rabbits of exogenous ketosis due to sodinm butyrate, the degree of ketosis became more pronounced either in normal or in alloxan animals than when sodium butyrate was given alone, especially, the elevation of β-hydroxybutyric acid was definitely marked. II Ketogenic activity of pituitary adrenocortical hormones for alloxan diabetic dogs.<BR>(1) When ACTH was administered for alloxan diabetic dog, the total blood ketone bodies increased markedly and both acetone and β-hydroxybutyric acid reached the maximum levels after one hour, and the former showed rapid depletion, while the latter showed gradual lowering.
