Kanamycin (20 mg/kg) was given intravenously to mongrel dogs, and changes in concentrations of the central and the peripheral compartments were calculated by means of pharmacokinetic analysis. Pancuronium bromide (0.02 mg/kg) was given intravenously every 90 minutes, then its effect on muscle relaxation was studied. The effect of pancuronium was prolonged, and its prolongation was most remarkable when the concentration of kanamycin in each compartment was decreased. <I>In vitro</I>, the contraction induced by acetylcholin was inhibited by pancuronium and kanamycin in an incubated solution, and it was observed that the concentration of kanamycin in the muscle remained in relatively high concentration after washing out the solution without kanamycin. It was thus postulated that muscle relaxation induced by pancuronium bromide was potentiated by kanamycin in the muscle, concentrationdependently in calculated compartments.
