Attempts have recently been made in many fields to apply flow cytometry measurement of DNA in tumor cells in the assessment of malignancy. In cancers of the head and neck, however, analysis of DNA histograms is complicated by the presence of various amounts of non-epithelial cells, including inflammatory cells. We theorized that the use of a monoclonal antibody for cytokeratin, a protein found i n the intermediate filament of epithelial cells, may make possible, the isolation of epithelial cells from tumor tissue, thereby enabling the determination of the DNA content of tumor cells. In the present study, we examined propidium iodide-fluorescein isothiocyanate-labelled-cytokeratin double staining by monoclonal antibody in KB cells. We succeeded in isolating KB cells from a mixture of KB cells and lymphocytes.
