It has previously been shown in the guinea pig that the negativity of the EP induced by systemic or perilymphatic application of the “loop” diuretics (such as ethacrynic acid, furosemide and bumetanide) is sharply reduced by ischemia or other types of anoxia.Recently Bosher [J. Physiol. 293 (1979)] reported that he could not detect the anoxia-sensitive negative EP following application of ethacrynic acid in the rat.In a systematic series of experiments we have found that this potential is present to a comparable degree in the rat and guinea pig, following both systemic and perilymphatic application of “loop” diuretics. Moreover the potential is present in chinchillas and dogs. The anoxia-sensitive negative EP thus appears to be a generalized response to “loop” diuretics in mammals.In addition we have found that this potential is also present following application of a new “loop” diuretic, piretanide.
