The antibacterial mechanism of p-aminosaltiric acid (PAS) has not been known up to present. While studying the biosynthesis of purine in M. tuberculosis, the author could clarify the antibacterial mechanism of PAS. The accumulation of 4-amino-5-imidazol-carboxamide (AICA), the intermediate of the biosynthesis of purine was recognized in the culture medium in which the growth of M. tuberculosis had been partially inhibited by means of PAS. Judging from the experimental results that the antibacterial action of PAS was inhibited by adenine, guanine, hypoxanthine and ribonucleic acid, it was inferred that PAS can inhibit the biosynthesis of purine through the inhibition of the ringclosing by putting one carbone into 4-amino-5-imidazol-carboxamide.
