For the purpose of evaluating the combined effects of 1-hexylcarbamoyl-5-fluorouracil (HCFU) and X-ray irradiation, the inhibition of isoproterenol (IPR) stimulated DNA synthesis in mice parotid glands and the survival time of mice bearing Ehrlich ascites tumors were used. When HCFU was given after irradiation, the combined effects were seen to be more potent than HCFU given before irradiation in both experiments. Especially when given at intervals of 2 hours after irradiation, the combined effect of HCFU (20mg/kg) and irradiation (400 rad) was found to have a synergistic effect on the mice bearing tumors and at least an additive combined effect on inhibiting IPR-stimulated DNA synthesis in the combination of HCFU (20mg/kg) with irradiation (100 rad). It was suggested that the synergistic effect can be expected in the clinical treatment of cancer, with 2 agents used in combination.
