The "fed-batch" culture method was used with the fungus Chaetomium gracile to produce endo-dextranase (EC 3. 2. 1. 11). The production process was divided into two stages: fungal growth and enzyme production. In the growth stage culture was with glucose as a carbon source, and in enzyme production feed-culture was with a minimum amount of dextran or its hydrolysate instead of glucose. During the enzyme production stage, the dissolved oxygen concentration was controlled between 1 and 5 ppm by the "On-Off" feeding of dextran. Dextranase productivity rose more than 12-fold by this new fed-batch culture method compared with the conventional culture method.
