The effects of ecdysterone and nerolidol on the incorporation of 14C-glycine into cultured posterior silk glands and cell-free system were examined. The posterior silk glands at 3 rd to 4 th day of the fifth instar were cultured in three mediums and a cell-free system which contained the com-bination of a small amount (10, 0.1, 0.001μg/m1) of ecdysterone and nerolidol. The enhancement of 14C-glycine incorporation up to 2 or 3 folds was observed when the glands were cultured in Wyatts medium without hemolymph. The enhancement became obscure when the glands were cultured in Wyatts medium with hemolymph. In the medium of Nakanishi et al. rich in nutrient, these two hormones showed an inhibitory effect. The hormones added to the cell-free system slightly affected the protein synthetic activity of this system. It seems likely that the enhancement of 14C-glycine incorporation is reflective of the increase of fibroin biosynthesis in cultured silk glands by a small amount of ecdysterone and nerolidol.
