A polysaccharide fraction, which, as determined by carbon clearance (C. C.) method, has a stimulative activity on phagocytosis in reticuloendothelial systems of rat, was found to exist in an aqueous extract of Chlorella cells. The acetone-defatted powder of the commercial Chlorella cells preparation was subjected to extraction at 80°C for 6 hr with water, and subsequent deprotein ization of the extract with basic lead acetate, followed by lyophilization, was carried out with 3 repetitions. Contrary to an unstable C. C. activity of the original aqueous extract, the crude extract preparation thus obtained, which was easily soluble in water but yet contaminated with nitrogenous matters (8.0% as N), indicated a stabilized activity. Gel filtration of this crude extract preparation with Sephadex G-75 gave two polysaccharide fractions; the first peak fraction, of which polysaccharide was mainly composed of rhamnose and galactose as hexose moiety, was of a fewer N content (2.2%) but of a feeble C. C. activity, while the second peak fraction, though contaminated with proteinous matters with rich N content (3.8_??_9.1%), exhibited a marked C. C. activity. The latter active fraction, of which polysaccharide was composed of glucose alone as main hexose moiety, was further deproteinized by pronase treatment and subsequent dialysis, which brought about a considerable decrease in N content (0.6_??_0.8%) as well as a marked elevation of sugar content (from 14.0% to 85.0%) together with an almost unchanged but rather enhanced C. C. activity. Thus, the active substance contained in Chlorella cells is evidently ascribed to their polyglucoside component. A presumptive estimation of molecular weight (MW) of polyglucoside contained in the active fraction, employing comparative gel filtration pattern method with Sephadex G-75, showed ca. 3500, based on the behavior of glucagon (MW 3500) as control.
