The synthesis of vitamin B1 (V. B1) by Saccharomyces saké, which was cultured in the READER's synthetic culture fluid, was tested with or without addition of pyrimidine and thiazole. It was found that the amount of V. B1 synthesised varied according to the kind of pyrimidine. The yield of V. B1 synthesised from 2-methyl-4-amino- 5-amino-methyl-pyrimidine and 4-methyl-5-β-hydroxymethyl-thiazole, which are the moieties of V. B1, in the culture fluid was about 100% or more than that of theoretical, but the yield from 2-methyl-4-hydroxy-5-hydroxymethyl-pyrimidine and thiazole was small, similar to the yield when only thiazole was used. The yeast containing larger amount of V. B1 was gotten, when the amounts of 2-methyl-4-amino-5-amino (or hdroxy)-methyl-pyrimidine and thiazole were increased more than before, but in this case the yield for theoretical decreased to about 43%.
