:Gastric Acid Secretion and Role of Cellular Cyclic AMP in Isolated Guinea Pig Gastric Glands

Isolated gastric galnds were prepared from guinea pig stomach by digestion with purified collagenase, in an attempt to determine gastric acid secretatory machanism at cellular level. Histamine increased significantly the production of cellular cyclic AMP and 14C-aminopyrine accumulation ratio in these glands. This action of histamine was inhibited by cimetidine (H2 antagonist), but not by mepyramine (H1 antagonist) except at very high doses. Thus, cyclic AMP seems to play an important role in the histamine induced acid secretion and histamine receptor can probably be classified as the H2type. In contrast to histamine, cellular cyclic AMP was not altered by pentagastrin nor carbachol. But, 14C-aminopyrine accumulation ratio was increased significantly by each stimulant. From these results, "three receptor theory"was supported rather than"histamine common final mediator theory"in the gastric acid secretatory mechanism.
