Scanning and transmission electron microscopic studies were performed on the cytotoxicity of the lymphocytes, from healthy human subjects or from patients with hepatoma, against human cultured hepatoma cells (LiC-1 Ohtsuka). The target cells were more strongly injured by the lymphocytes from healthy subjects, especially by non-E rosette forming cells with many microvilli, than by those from patients with hepatoma.Close contact between the lymphocytes and the target hepatoma cells was considered to be essential for target cell injury. At the initial stage of the mixed culture, the microvilli or the uropod projected from the lymphocyte surface were intimately connected with the microvilli on the target cell (interdigitation). After 24 hour incubation, more broad contact was seen between the plasma membranes of both cells (peripolesis), concomitant with many blebs on the target cell, indicating the degeneration of the cell. The plasma membrane of the lymphocyte deeply brought into the target cell like a pseudoinclusion body was entirely attached to that of the target cell (emperipolesis). In this form of cell contact, most prominant ultrastructural alterations, i.e. an increase in autophagic vacuoles and lysosomes and large bleb formation, were observed in the target cell. At the terminal stage, the target cell was found to be in a lytic necrosis. At high magnification of the cell contact area, the narrow gap (200-400 Å in width) was always present between the opposed plasma membranes. The intercellular gap, however, was filled with the ruthenium redpositive surface coats on the outer surfaces of both plasma membranes, which were closely associated or fused each other. The lymphocytes may affect the target hepatoma cell through the connection of both cell surface coats.
