Effects of Anticancer Drugs on Protein Synthesis of the Exocrine Pancreas

In an attempt to evaluate effects of anticancer drugs on protein synthesis of the exocrine pancreas, 5-Fluorouracil (10-30mg/kg body weight), Actinomycin D (0.025mg/kg body weight), Vincristine (0.25mg/kg body weight), Methotrexate (0.4mg/kg body weight) or Daunomycin (1.5mg/kg body weight) were administered intraperitoneally to male Donryu rats on fasting once daily for 3 days. Uptakes of labelled amino acids (75Se-Selenomethionine, 3H-Leucine and 3H-Methionine), autoradiography and histology of the pancreas were examined.The following resalts were obtained.1) All anticancer drugs produced a decrease of75Se-Selenomethionine uptake to the pancreas. This effect was particulary prominent with 5-Fluorouracil and Vincristine.2) 3H-Leucine uptake to the pancreatic protein fraction (TCA-insoluble fraction of pancreas homogenate) was diminished with 5-Fluorouracil and Vincristine to 72% and 57%, respectively, of control levels.3) Autoradiography demonstrated a decrease in the amount of grain on zymogen granules in cases treated with 5-Fluorouracil, Vincristine or Actinomycin D in comparison with nontreated controls.4) Histological examination revealed the enlargement of cells and increase in number of zymogen granules in the caces treated with 5-Fluorouracil, while it showed the shrinkage in size of the cells and diminition of zymogen granules in the cases treated with Actinomycin D or Vincristine.
