• 1) The metabolism of endogenous and exogenous urinary cortisol by means of the administration of cortisol-4-<SUP>14</SUP>C or cortisone-4-<SUP>14</SUP>C was studied in normalsubjects, patients with hyperthyroidism and Cushing's syndrome.<BR>2) The urinary outputs of THE and ATHF in hyperthyroid patients were increased above the level in normal subjects, while the output of THF was similar to normal subjects. The ratios of ATHF/THE and THE/THF+ATHF in hyperthyroidism were increased about 3.0 and 2.0 times more than those in normal subjects. It would appear that in hyperthroidism 5α-reduction of A ring of steroid is predominant over 5β-reduction and that the increased THE is due not only to an increased 11-oxidation (F→E), but to an increased reduction of A ring (E→THE).<BR>3) The urinary output of THF in Cushing's syndrome on the other hand, was increased in comparison with that of THE. The ratio of THE/THF+ATHF was decreased to about 50% of that in normal subjects, while the ratio of ATHF/THF was similar to that of normal subjects.
