• <I>A total synthesis of (+) -leinamycin (1), a unique sulfur-containing antitumor antibiotic, is described. The present synthesis features a stereocontrolled construction of the macrolactam followed by a spiroannulation of the hitherto unprecedented dithiolanone ring.</I>
