For removing the membrane of dog salmon egg a single treatment of it with proteolytic enzymes is not sufficient. Enzymatic dissolution of the egg membrane is attained only for the eggs that have previously been treated with acid solution. The most suitable procedure for removing the egg membrane is composed of a series of treatments as follows: 15 minutes immersion in acidulated RINGER'S solution (pH 1.8) → washing with normal RINGER'S solution → immersion in 0.2% pancreatin-RINGER'S solution (pH 11.0). The egg thus denuded is still fertilizable. Pancreatin enzymes effective for dissolving the egg membrane are likely to be tryptic enzyme and cathepsin. The double treatment with acid and pancreatin solution induces egg activation without breakdown of cortical alveoli. About this phenomena some discussion is presented.
