Multicellular rat hepatocyte spheroids were primary cultured on a hydrophobic plastic dish and cell organization and reconstitution of cell polarity therein were studied. As culture time elapsed, hepatocytes aggregated forming spheroids in which bile canaliculi and junctional complexs reappeared. K<SUP>+</SUP>-dependent p-NPPase, Mg<SUP>2+</SUP>-ATPase and ALPase activities also showed supporting facts for reorganization of hepatocytes and reconstitution of cell polarity through appearance and disappearance of thier activities each. Lanthanum nitrate and cationized ferritin used as tracers also confirmed the reorganization of cultured hepatocytes. The results suggested that cellular reorganization and the maintenance of reconstituted cell polarity nearly as in the in vivo state were carried out better and longer in the conventional monolayer culture.
