Expression of interleukin 2 and interleukin 2 receptor (p55) genes in PHA and TPA-stimulated peripheral blood mononuclear cells from HBV carriers.

In order to clarify the pathogenic role of interleukin 2 (IL2) and IL2 receptor (IL2R) system in HBV carrier state, expression of IL2 and IL2R gene were investigated using Northern blot hybridization in cultured peripheral blood mononuclear cells stimulated with phytohemagglutinin (PHA) and 12-O-tetradecanoyl-phorbol-13-acetate (TPA). The amounts of IL2 mRNA were found to be decreased in 8 out of 15 patients with type B chronic active hepatitis (CAH) and in 6 out of 8 asymptomatic HBV carriers (ASC). The expression of IL2R gene was also decreased in a part of patients with decreased IL2 mRNA levels, while mRNA levels of c-<I>myc</I> gene were almost the same in all patients. Furthermore, IL2 concentration in culture supernatant of peripheral blood mononuclear cells from patients with type B CAH was significantly lower than that from normal subjects, suggesting that abnormality of IL2 and IL2R gene expression was related to HBV carrier state.
