This study was performed to examine the localization of 2', 5'-oligoadenylate (2-5A) and the antiviral status in the liver using indirect immunoenzymatic technique and double staining. 2-5A in the liver was observed in the cytoplasm of hepatocytes of 42 out of 46 patients with viral hepatitis. In chronic hepatitis type B, hepatocyte 2-5A was stained more strongly 2-5A during the increasing phase of DNA-polymerase activity than during the peak or decreasing phase (p<0.05).<BR>Under the immune electron microscopy, 2-5A was found in the cytosol of cytoplasmic matrix, but not in the nucleus.<BR>In double staining, the simultaneous presence of HBeAg was significantly lower in hepatocytes strongly staining for 2-5A (p<0.001). This indicates that 2-5A inhibits viral replication.<BR>By interferon (IFN) treatment, the increase of 2-5A in hepatocytes was found in 7 out of 11 patients with chronic active hepatitis type B. In 3 of 7 patients with increased 2-5A, their sera were found to have converted to HBeAg negative.<BR>These results indicate that 2-5A plays a role in antiviral activity in situ.
