C型慢性肝炎における肝局所のサイトカイン mRNA の発現と臨床病理学的所見との相関に関する研究

The author investigated the relationship between mRNA expressions of cytokines and liver biopsy findings in chronic hepatitis C (CH-C) by reverse-transcription polymerase-chain-reaction. The mRNAs of interleukin-1α, interleukin-2, interleukin-6 and tumor-necrosis-factor-α were not detected in any of the patients with and without CH-C. The mRNAs of interleukin-2 receptor and interferon-γ were detected in most patients with CH-C, but not in any of the patients without CH-C. The mRNAs of interleukin-1β and interleukin-8 were detected in most patients with and without CH-C. Transforming-growth-factor-β<SUB>1</SUB> mRNA showed strong expression in all patients. The intensity of interleukin-8 gene expression was correlated with the total score of histological activity index, and especially with the score of portal inflammation and periportal+/- bridging necrosis. These findings suggest that the upregulation of gene expressions in interleukin-2-receptor, interferon-γ and interleukin-8 may be associated with the pathogenesis of CH-C and interleukin-8 especially associated with the histological activity of CH-C.
