Immunohistochemical study of expression of P-glycoprotein in hepatocellular carcinoma.

Expression of P-glycoprotein in fifteen patients with hepatocellular carcinoma were investigated by immunohistochemical study using anti-P-glycoprotein monoclonal antibody (C219, Centocor, Inc, USA). P-glycoprotein was demonstrated on biliary canalicular surface of tumor in hepatocellularcarcinoma (HCC), as similar as in normal hepatocytes. P-glycoprotein labeling indices (P.L.I.) were as follows: 0.80±0.12 (Mean±SD)in non-cancerous liver tissue, 0.46±0.13 in moderately differentiated HCC and 0.04±0.05 in poorly differentiated HCC. Expression of P-glycoprotein was dependent on histological differentiation of HCC.<BR>These results suggest that expression of P-glycoprotein in non-cancerous and tumors is related to sensitivity for anti-cancer chemotherapy and it's remarkable expression in normal hepatocytes may explain a mechanism of low sensitivity to anti-cancer agents in normal hepatocytes. Expression of P-glycoprotein in hepatic cancer might be an appropriate index of sensitivity to anti-cancer chemotherapy.
