The activity of sterol-ester hydrolase (E.C. 3. 1. 1. 13) has been determined in various tissues such as rat liver. ratpancreas, rat brain, human placenta and human liver. This enzyme hydrolyzes esterified cholesterol to free cholesterol and seems to play an important role in cholesterol metabolism. However, the physiological significance of this enzyme has not bcen demonstrated.<BR>In tbese cxperiments, the optimum pH for the hydrolysis of [4-<SUP>14</SUP>C] cholesteryl oleate was investigated in rat liver homogenates, and also in the micmsomal and supernatant fractions. In addition, alteration of sterol-ester hydrolase activity following the experimental liver injury induced by CCI<SUB>4</SUB> and biliary obstruction was determined.<BR>The optimum pH for enzyme activity was found over a broad range between 6.6 and 7.4 in the liver homogenates, 7.0 in the microsomal fraction and 7.8 in the supernatant fraction.<BR>In the experimental liver injury, the activity of sterol-ester hydrolase increascd significantly in the microsomal fraction, however, was within normal levels or less in the supernatant fraction.<BR>In rat serum, the activity of sterol-ester hydrolase was present in very low levels in control and also in the experimeDtal liver injury.
