The Peroxidase anti-peroxidase complex method with rabbit anti-hCG antibody as the first antibody was used to demonstrated an hCG-like material in 9/46 (19.6%) of the tissue specimens of uterine cervical squamous cell carcinoma. A cervical epidermoid carcinoma cell line, SKG-I, was found to produce human chorionic gonadotropin β-subunit (hCG-β), but no distinct hCG. Treatment of SKG-I cells with sodium butyrate increased hCG and hCG-β immunoactivities.Comparative studies between standard urinary hCG-β and SKG-I producing hCG-β (SKG-β) were carried out by means of isoelectrofocusing (IEF) and lectin chromatographies. The standard hCG-β had 4 distinct components between pH 3.0 and pH 4.5. The SKG-β had 3 major peaks at pH 4.0 (SKG-β-I), pH 5.2 (SKG-β-II), pH 6.0 (SKG-β-III) and 1 minor peak at pH 10.1 (SKG-β-IV). The later 3 peaks were noted only in SKG-β. Lectin chromatographies (WGA and RCA-I) of standard hCG-β and SKG-β suggested that SKG-β possessed less terminal sialic acid than pregnancy urinary hCG-β especially in SKG-β-III and SKG-β-IV, and that the heterogeneity of SKG-β in IEF could be partly explained by the differences in the content of terminal sialic acid.
