The effects of NaNO<SUB>2</SUB> and NH<SUB>2</SUB>OH on mitochondrial MAO from rat brain were studied with the following results: NaNO<SUB>2</SUB> and NH<SUB>2</SUB>OH activated the enzyme. The effects of NaNO<SUB>2</SUB> and NH<SUB>2</SUB>OH on enzyme activity differed with different substrates. The activating effects of NaNO<SUB>2</SUB> and NH<SUB>2</SUB>OH on MAO activity were reversible. NaNO<SUB>2</SUB> did not cause activation at any concentration of pheniprazine and pargyline tested. However, 1×10<SUP>-2</SUP>M NH<SUB>2</SUB>OH decreased the inhibitions by pheniprazine and pargyline at all concentrations of the inhibitors tested. Addition of 1×10<SUP>-2</SUP>M NH<SUB>2</SUB>OH changed the Km value of MAO for tyramine from 1.25×10<SUP>-2</SUP>M to 6.2×10<SUP>-3</SUP>M.
