Experimental Study on the Effect of Sclerosing Agents for Hemorrhoids.

In Japan, 5% phenol almond oil(PAO) is clinically used as a sclerosing agent for internal hemorrhoids. However, the effect is neither permanent nor remarkable for prolapsing hemorrhoids. The development of a new drug with the better effects is expected. In our aminal experiments, PAO, 5% ethanolamine oleate(EO), 1% polidocanol (PO), and OC-108, which are being researched in our country, were injected into the rectal submucosa of rats. The sclerosing effects of these 4 drugs on necrosis, edema, cellular infiltration, ulcer formation, new vascular formation, fibroblast, and fibrosis were microscopically observed for 21 days. Although the results show no significant differrence in these histological items, the only peculiar finding is granuloma formation in the specimen obtained from 21 days to 9 weeks after injection of OC-108. The granuloma can change to cicatrix which is considered to have a prolonged sclerosing effect.
