Overexpression of p 53 was studied immunohistochemically in 124 colorectal cancers. Cellular DNA ploidy pattern of 97 colorectal cancers were investigated by using a microspectrophotometer with DAPI staining. These results were evaluated with clinicopathological findings, prognosis, and the positive rate of PCNA immunohistochemical staining.<BR>The overexpression of p 53 was recognized in 52 carcinomas (41.9%) . It showed only significant correlation with factor v of pathological findings (p<0.05 ). The tendency of recurrent pattern in the p53 positve cases was peritoneal dessemination.<BR>In DNA ploidy pattern, the aneuploidy had more advanced cases and there was significant correlation between aneuploid cancers and positivity of ly, n factors, and hepatic metastasis (p <0.01, p <0.05, and p<0.05, respectively). However, DNA ploidy pattern was unrelated to the positive rate of PCNA. On five-year survival rate, aneuploidy cases were significantly worse than those in diploidy (p<0.05). On the other hand, there was no significant difference when the analysis was limited to the cases of curative resections. In recurrent pattern, aneuploid cancers tended to occur hematogenous metastasis whereas diploid cancers more often demonstrated peritoneal dissemination (p<0.05).<BR>These results suggest that DNA ploidy pattern serve as a useful prognostic indicator in colorectal cancer.
