Evaluation of DNA ploidy and DNA indices with prognosis of colorectal cancer by FCM.

We studied the association of DNA indices (DIs) with clinicopathological factors and prognosis in 201 patients with primary colorectal cancer by flow cytometry, using paraffin-em-bedded tissue. One hundred and eight (53.7 %) tumors showed aneuploid.<BR>Aneuploid tumors were classified into three groups, 47 tumors with DI from 1.01 to 1.5, 47 with DI from 1.51 to 2 and 14 with DI more than 2. DNA ploidy (diploid/aneuploid) correlated significantly with location (colon/rectum) and lymphatic invasion (-/+). On the other hand, DIs did not correlate with any other factors. Patients with aneuploid tumor had significantly worse prognosis than those with diploid tumors in all cases and in stage IV. In a Cox multivariate regression analysis, histological stage, macroscopic type, location, lymphatic invasion and DI were powerful prognostic indicators in colorectal cancer. However, DNA ploidy was not recognized as prognostic indicator.<BR>These results suggest that DI rather than DNA ploidy was an important prognostic indicator in clorectal carcinoma.
