Tail sheath protein of phage T4 constitutes the outer cylinder of the co-cylindrical structure of the tail, which appears to have evolved for efficient infection of the phage.<BR>It consists of 144 protomer molecules arranged in 24 annuli. Contraction of the sheath is exothermic and irreversible. Recently, primary structure of the protomer tail sheath protein (gp18) was determined from nucleotide sequence of the gene, which was subsequently confirmed by peptide analysis of the gene product. Use of the primary structure and available gene 18 mutants for further studying structure-function relation of the gene product and the mechanism of sheath contraction are discussed.
