Four chlorophyll mutant strains of sand oats (albino-R5, -R6, R7 and yellow-1) and two of barley (yellow chlorina and yellow green) were cultured in test tubes to investigate their nutritional requirements for chlorophyll formation. Plants were cultured in test tubes containing nutrient agar media. Effects of glucose, casein hydrolysate, and yeast extract on chlorophyll formation were examined using leaves of these mutants. Glucose was effective on albino-R5 and yellow-1 mutants of oats to increase chlorophyll content, while casein hydrolysate and yeast extract were ineffective. In albino-R6 and -R7 no increase of chlorophyll was observed in the media used. In yellow chlorina and yellow green mutants of barley, the addition of glucose caused an increase of chlorophyll content. Only yellow green mutant responded to casein hydrolysate, whose increasing concentrations resulted in a higher content of chlorophyll. Alanine, valine, leucine and glutamic acid also stimulated chlorophyll formation in this mutant.
