Dynamic Changes in Histone Acetylation during Sheep Oocyte Maturation

The changes in histone acetylation are not always consistent in various cell types and at different developmental stages. To understand the detailed dynamic changes in histone acetylation during sheep oocyte meiosis, we performed the immunostaining of specific antibodies against acetylated lysine 9 on histone H3 and acetylated lysines 5, 12 on histone H4. We found that the acetylation fluorescence signals of H3/K9 and H4/K12 on chromatin appeared intensively at germinal vesicle (GV), late-GV (L-GV) and germinal vesicle breakdown (GVBD) stages, and became weak at metaphase I (MI), but staining reappeared at anaphase I-telophase-I (AI-TI) and metaphase II (MII). Furthermore, the staining was detected in the first polar bodies. The fluorescence signals of H4/K5 appeared firstly at MI stage, became intensive at AI-TI stage, but was barely detectable at MII stage chromosomes and first polar bodies. Together, we conclude that H3/K9 and H4/K12 have similar acetylation pattern, while H4/K5 has its unique acetylation pattern during sheep oocyte meiotic maturation.
