Phenoloxidase (Diphenoloxidase EC 1.10.3.1) was purified from an aqueous extract of the larvae of the housefly Musca domestica vicina Maquart by a procedure involving affinity chromatography on a Sepharose 4B derivative of p-aminobenzoic acid and ethylene diamine, Sepharose 4B and Sephadex G-200 gel filtrations. The purified phenoloxidase was homogeneous on polyacrylamide gel electrophoresis. By this improved method, phenoloxidase was recovered in greater yield and specific activity as compared with our previous purification procedure.
