A new technologies are required for rapid surveillance of the current highly pathogenic H5N1 avian influenza A circulations to survey the potential for adaptation of the virus to the human type receptor, an important step in the emergence of pandemic influenza virus strains. Hemagglutinins (HAs) from avian and human viruses are characterized by their preference for α2-3 (Avian type receptor: Neu5Acα2-3Galβ1-) and α2-6-linked N-acetylneuraminic acid (Neu5Ac) (Human type receptor: Neu5Acα2-6Galβ1-), respectively. Owing to the species-specific nature of the interaction between the virus and host glycans, attention has focused on novel immunochromatography technologies that can rapidly assess virus receptor specificity with high sensitivity and the potential emergence of human type receptor-adapted H5N1 viruses. We have developed an advanced prototype of immunochromatography-based device to differentiate H5N1 viruses that bind to avian- and human-type receptors. This procedure enables us high-sensitivity detection of sialo-glycan binding of avian and human influenza viruses (4HAU-128 HAU). This technology allows investigation and surveillance of mutated avian influenza viruses that acquired the adaptation to the human type receptors (Neu5Acα2-6Galβ1-) on single strip to which different receptor sialo glycan structures can be coupled.
