Effects of various divalent cations, such as Ni++, Co++, Zn++, Mn++, Cd++, Hg++, Sr++, Ba++ and trivalent cation La+++, on both ATPase activity and Ca binding of intestinal myosin B were observed.The results obtained are as follows; 1) Sr++ and Ba++ which have ionic radii larger than 11.5 Å exerted no effect on the Ca binding activity, but activated the ATPase activity of myosin B by about 20%. 2) Ni++ which has a smaller ionic radius than 0.7 Å inhibited Ca binding activity by a few percent, and inhibited ATPase activity by 55%. 3) Co++, Zn++, Mn++, Cd++ and Hg++, whose ionic radii are between 0.7 and 1.1 Å, inhibited the ATPase activity in proportion to their inhibition of the Ca binding activity respectively. While La+++ completely inhibited the ATPase activity of myosin B, it depressed Ca binding by 55%. Based on these results, the dissimilarity of actomyosin extracted from both skeletal and smooth muscle with regard to the influence of various divalent and trivalent cations was discussed.
