In order to investigate the effect of insulin degradation products upon the biological activity of insulin against tissues, discrepancy of insulin activities in serum between immunoreactive insulin (IRI) measured by radioimmunoassay and insulin-like activity (ILA) measured by biological methods was studied clinically, and the relation of insulin degradation product to the discrepancy between IRI and ILA was studied in animal experiment. The results obtained were as follows : 1. Values of ILA in sera measured by rat diaphragm (DILA) and ILA in sera measured by rat epididymal fat pad (FILA) were far higher than the values of IRI performed by parallel determination in the same serum samples. Values of DILA in sera of the patients with diabetes mellitus were lower than that of healthy subjects, in contrast, values of FILA in diabetics were higher than that of healthy subjects. 2. Degradation of insulin by diaphragm tissue or epididymal fat tissue was investigated using 131I-labeled insulin and thin-layer chromatographic analysis. The ratio of insulin degradation by tissue was lower in the case of insulin in serum than that insulin in buffer solution. The finding indicates that DILA in serum is expressed as an approximately 2 fold concentration, and FILA is expressed as an approximately 1.6 fold concentration by calculation using the insulin standard curve depicted by a system of the buffer solution. This could be one of the causes of the discrepancy of insulin activities between IRI and ILA in serum. 3. Insulin degradation products, induced by muscle tissue, inhibited the insulin degrading enzyme activity and also reduced to some extent the biological effect of insulin on the diaphragm. The fact suggests that the degradation product may be competitive with insulin against the receptor of muscle cell membrane. Insulin degradation products, on the other hand, slightly accelerated the biological effect of insulin on the fat tissue. This suggests that the degradation product may not be competitive with insulin, but may rather be antagonistic with insulin with respect to receptor of fat cell membrane.
